It arrived in Australian conversation the way most Korean innovations do — quietly, through clinics, several years after it had already become standard practice in Seoul. Here is what polydeoxyribonucleotide actually is, and what it can honestly do for your skin.

If you have had a facial in the last eighteen months, someone has probably said the word PDRN to you. It is on menus in Double Bay and South Yarra, in the after-care instructions of injectors who do not put anything on a menu without reason, and increasingly on the boxes of products sitting in the bathrooms of women who pay close attention.

It is also, unhelpfully, becoming a marketing word. Which is the moment we usually stop and write something like this.

What PDRN actually is

PDRN stands for polydeoxyribonucleotide. It is a fragment of DNA — specifically, a chain of purified, sterilised DNA fragments, most commonly derived from the sperm cells of chum salmon.

That sentence tends to stop people, so let us stay with it for a moment. Salmon DNA is used because it is remarkably compatible with human DNA at the sequence level — high enough homology that the human body reads the fragments as familiar rather than foreign. It is abundant, it is a by-product of an existing food industry rather than a purpose-farmed material, and it can be purified to a pharmaceutical standard. Those three facts are why it became the source material and not something more glamorous.

The fragments are not intact DNA in any meaningful sense. They are short chains, in the region of 50 to 1,500 kilodaltons, deliberately broken down to a molecular size that can interact with cells rather than simply sit on top of them.

The mechanism, in plain terms

PDRN works primarily through what is called the adenosine A2A receptor pathway. When the fragments break down at the cell surface, they release nucleotides — the building blocks of DNA — which bind to adenosine receptors on fibroblasts. Fibroblasts are the cells in your dermis responsible for producing collagen, elastin and hyaluronic acid. They are, functionally, the cells responsible for your skin being firm.

Activating that receptor does two things at once. It signals fibroblasts to increase collagen synthesis, and it quiets inflammatory signalling. That combination — build more, inflame less — is unusual. Most actives that stimulate the dermis do so through controlled irritation. Retinoids work partly by provocation. Acids work by removal. PDRN is one of the very few categories that asks the skin to build without first asking it to be injured.

There is a second, more elegant mechanism running alongside it. The released nucleotides can be recycled directly into the cell’s own DNA repair processes — the so-called salvage pathway. In effect, you are handing a cell the raw materials for repair at the moment you are also telling it to repair. It is a rare piece of biological tidiness.

Why it emerged from Korea, and why it took so long to reach us

PDRN has a longer medical history than its beauty-counter debut suggests. It has been used in Italy and Korea for decades in wound healing, diabetic ulcer management and post-surgical tissue repair. Its migration into aesthetics followed the ordinary route: surgeons noticed that treated tissue healed better and looked better, and the aesthetic industry followed the surgeons.

Korea then did what Korea does exceptionally well. It industrialised the ingredient, refined the extraction, drove the cost down, and built an entire treatment category around injectable PDRN — the treatment most Australians know by the brand name that dominates the category rather than by the molecule.

The topical formats came later, and they came with a legitimate scientific problem attached.

The honest limitation: molecular size

PDRN is a large molecule. The skin barrier is very good at excluding large molecules — that is, after all, its entire professional purpose. So the obvious question, and the one most brands would rather you did not ask, is how much of the PDRN in a topical product reaches the dermis where fibroblasts live.

The honest answer is: less than an injection delivers. Considerably less. Any brand telling you otherwise is selling you something.

The more useful answer is that this does not make topical PDRN pointless, for three reasons.

  1. Formulators use low-molecular-weight PDRN fractions specifically selected for penetration, rather than the full-size molecule used in injectables.

  2. Delivery format matters enormously. An occlusive sheet mask held against the skin for twenty minutes creates hydration, warmth and a sealed environment — conditions under which the barrier becomes measurably more permeable than it is in open air. A balm applied to warmed skin behaves similarly.

  3. The epidermis is not a passive bystander. Keratinocytes carry adenosine receptors too. Activity at the upper layers has real consequences for barrier quality, redness and the way light reflects off the skin surface.

So the correct expectation for a topical PDRN product is not "an injection in a jar". It is: better-behaved skin, faster recovery from irritation, visibly improved surface quality, and — with consistent use over months rather than weeks — a contribution to the slow work of dermal maintenance.

Who it suits

PDRN is the rare active we recommend for skin that cannot tolerate much. Because it is anti-inflammatory rather than provocative, it sits comfortably alongside compromised barriers, rosacea-prone complexions and post-procedure skin. It is the ingredient we would reach for in the week after a laser or a peel, not the week before.

It is also, for our reader specifically, a sensible answer to a very common midlife problem: skin that has become reactive at precisely the moment it also needs stimulation. PDRN is one of the few ways to do both at once.

How to actually use it

  • Apply to slightly damp, clean skin. Hydration improves penetration of everything, and PDRN more than most.

  • Use it before your heavier occlusives, not after. Nothing gets through a shea-based balm.

  • Consistency beats concentration. A weekly mask over four months will outperform a fortnight of enthusiasm.

  • Do not layer it with a strong acid or retinoid in the same application. Not because of a reaction, but because you have just paid for an anti-inflammatory and then immediately inflamed the skin.
